RESUMO

While the passive transfer of immunity in horse and donkey foals has been extensively studied, there is limited information for mule foals. Immunoglobulin type G (IgG) and serum total protein concentration (TP) were assessed at different sampling times to evaluate the correlation between serum radial immunodiffusion (SRID) with electrophoresis, refractometry, and dry chemistry analyzer (Biuret), and to estimate serum IgG concentrations using serum TP in mule foals. We analyzed a total of 30 samples collected at birth, and at 6, 12, 24, and 48 h of life from 6 mule foals by SRID, electrophoresis TP, biuret TP, and refractometry TP. The SRID IgG concentration significantly increased from birth until T6 (p < 0.001). Serum TP analyzed with refractometry revealed differences between T0 and T12, T24 and T48 (p < 0.05), while a significant difference was observed with the biuret method between T0 and all the other sampling times (p < 0.001). A strong correlation was found between IgG SRID and biuret TP (r = 0.69, p < 0.001), and a good correlation existed between IgG SRID, refractometry TP, and electrophoresis TP (r = 0.44, p < 0.01 and r = 0.39, p < 0.05, respectively). All methods can be used to estimate the passive transfer of immunity in mule foals. TP refractometry and biuret TP values can be used to determine serum IgG concentrations in the blood of mule foals on their first day of life through the application of a specific equation.

Background@#Homocysteine (HCY) was evaluated in healthy and chronic enteropathic dogs, however no studies on dogs with immunosuppressant-responsive enteropathy are available. @*Objectives@#The aim was to evaluate serum HCY concentrations and its prognostic role in dogs with immunosuppressant-responsive enteropathy compared to healthy dogs. @*Methods@#Serum HCY concentration was statistically compared between 24 healthy dogs and 29 dogs with immunosuppressant-responsive enteropathy. Correlation analyses between serum total protein, albumin (ALB), C-reactive protein (CRP), folate and cobalamin, and serum HCY concentration were performed in immunosuppressant-responsive enteropathic dogs. @*Results@#The associations between serum HCY concentration and clinical, histological, endoscopic scores and follow-up were evaluated. Mean serum HCY concentration was higher in immunosuppressant-responsive enteropathic dogs compared to control dogs (30.22 ± 8.67 μmol/L vs. 5.26 ± 2.78 μmol/L; p < 0.0001). No association between serum HCY concentration and total protein, ALB, CRP, folate concentration as well as, clinical score, histological and endoscopic scores was found. A negative correlation between serum HCY concentration and cobalamin was noted (p = 0.0025, r = −0.54). No significant difference in HCY was found between responsive and non-responsive dogs or between survivors and non-survivors. @*Conclusions@#Although, serum HCY concentration was higher in immunosuppressant-responsive enteropathy, its prognostic value remains unclear. However, further prospective, large-scale studies are warranted to better investigate the possible prognostic role of HCY in immunosuppressant-responsive enteropathic dogs.

We present a new method for the rapid analysis of ochratoxin A (OTA) in pig tissues (muscle, liver and kidney) using enzymatic digestion (ED) coupled to high-performance liquid chromatography with a fluorescence detector (HPLC-FLD). OTA was digested with a 1% pancreatin solution in a phosphate buffer and then cleaned with ethylacetate. After being evaporated to dryness and re-dissolved, the sample was determined using HPLC-FLD. The method was validated taking into account the currently permitted limit of 1 µg/kg OTA in pork meat and derived products in Italy. The recovery was higher than 90%. Intra- and inter-day repeatability expressed as RSD were less than 7%. The LOD and LOQ were 0.001 and 0.002 µg/kg, respectively. Our method is more efficient, easier, and cheaper than conventional clean-up procedures (liquid-liquid extraction). •The aim of the study was to develop and validate a quantitative HPLC-FLD method based on ED followed by a chromatographic analysis without any previous clean-up or concentration step for the detection of OTA in pig tissues.•The ED method showed a 90%+ recovery, and intra- and inter-day RSD less than 7%.•This method is simple, rapid, easy to use, and consumes low amounts of organic solvents.

Glomerular filtration rate (GFR) is still a highly underutilized tool in cats because available methods are not easy to be performed in clinical practice. Iohexol (IOX) has been shown to be a useful and reliable marker of GFR both in animals and in humans. The aim of the present study was to develop a rapid and reliable method for measuring IOX in feline plasma and to evaluate the accuracy of limited sampling models to establish a low-cost and clinically suitable GFR test. IOX concentrations were determined by using a new HPLC-UV method. GFR was assessed as plasma clearance of IOX, which was calculated by dividing dose administered by area under the curve of plasmatic concentration versus time (AUC), and indexed to body weight (BW). Correlation and agreement analysis between the GFR values obtained by a seven-point clearance method and the GFR values determined by the application of simplified sample combinations indicated that the 3-blood sample clearance model (5, 30, and 60 min) was the best simplified method because it provided an accurate GFR value in only one hour. The reported method is a simple and accurate way of GFR determination, which may be easily used in a clinical setting.
